Since you didn't specify the type or model of camera you're using, I can't be completely specific.

You can use any exposure mode you find appropriate. Auto usually works well, at least until the frame is dominated by dark leaves or a bright flower. If the scene is not midrange, you can either use exposure compensation or switch to manual exposure. If you're shooting digital, you can review the pictures, and use the histogram if available.

If you're using an SLR, you'll need a macro lens. Or extension tubes between the lens and body. Or a close-up adapters in front of the lens.

If you're using a point&shoot or super-zoom, you'll probably need to switch to a close-up or macro mode.

In any event, camera motion becomes critical for close-up work, as the short working distance magnifies every bit of camera shake. You're absolutely going to need a steady support. A tripod is best, but you might be able to get by with a beanbag, depending on your shooting conditions. You can also reduce camera shake by using the self-timer, so any motion caused by your trigger finger can damp out before the picture is taken. If your SLR has either a mirror lock-up mode or an exposure delay mode, you can use that as well to reduce the motion caused by "mirror slap."

Lighting may also become a concern. Make sure neither your camera nor your body cast shadows on the subject (unless you want to). You may want to consider using flash. If you're using a SLR, make sure you remove the lens hood if you use the built-in flash, otherwise you'll see the shadow cast by the hood.
