I have a Nikon D40, and when I set the mode dial to Close up the shutter button won't always depress.
It's because you are to close to the subject. You can back off or set your lens to manual and the shutter will release whether or not it is in focus.

Command dial on a friend's Fuji Finepix S9000 is NOT functioning. The shutter release button on my Nikon MB-D10 doesn't appear to be working. The switch on the side is turned to the dot as it should be.
